Comic book shops are generally the only "brick-and-mortar" place to purchase single issues.

Ebay is a good place to find single issues online.Book shops
Most larger book stores will stock graphic novels and trade paperbacks but generally won't stock single issues. I tend to find that bookshops are more expensive than anywhere else, but you can sometimes find a bargain! Note that their online stores will often have different prices than in-store.

The Comixology store sells individual issues (as well as all other formats), whereas Kindle store tends only to sell collected editions and graphic novels. DRM-Free Digital Comics Reading Methods
(eg .PDF or .CBR format reader)
DRM-free means that you are given the comic as a file to download without any restrictions (think of them as .mp3 files, but for comics). DRM-free comics can be obtained from many places, such as directly from the publisher, from other 3rd party storefronts, Humble Bundles, etc.
Certain publishers provide DRM-free copies of their titles via Comixology/Amazon.
